Apexbio Technology LLC Annexin V-Cy5/PI Apoptosis Kit 100assays
Annexin V is a Ca2 -dependent phospholipid-binding protein that specifically binds to phosphatidylserine (PS) Under normal conditions PS resides on the inner side of the cell membrane lipid bilayer However in early apoptosis PS translocates from the inner leaflet of the plasma membrane to the outer surface becoming exposed externally Annexin V exhibits high binding affinity for PS and when conjugated with fluorescent molecules (Cy5) it serves as a fluorescent probe to detect apoptosis using flow cytometry or fluorescence microscopy The staining procedure requires only 10 20 minutes PI specifically binds to double-stranded DNA to produce red fluorescence it is normally impermeant to intact cell membranes However during late-stage apoptosis or in dead cells PI penetrates the membrane to stain nuclear DNA Combined Annexin V-Cy5 and PI staining allows distinction between cells at various stages of apoptosis and dead cells

Annexin V is a Ca2 -dependent phospholipid-binding protein that specifically binds to phosphatidylserine (PS) Normally PS is localized on the inner leaflet of the cell membrane lipid bilayer During the early stages of apoptosis PS translocates from the inner leaflet to the outer surface of the cell membrane exposing it to the extracellular environment Due to Annexin V s high affinity for PS fluorescently labeled Annexin V (Cy5) can be used as a fluorescent probe to detect apoptotic cells through flow cytometry or fluorescence microscopy with staining taking only 10-20 minutes DAPI specifically binds to double-stranded DNA emitting blue fluorescence but normally cannot permeate intact cell membranes However DAPI can enter late-stage apoptotic or dead cells and stain their nuclei Simultaneous staining with Annexin V-Cy5 and DAPI allows differentiation of cells at various stages of apoptosis and cell death
